FIRST DAY: The Story Begin


WHO IS MR. FERMENTER?



Date : 4 October 2010
Time : 12.00 pm
Venue : Laboratory 148

We are quite excited to know and to work with our precious Mr . Fermentor named MINIFORS. This is the story 2 days before we are starting to use the bioreactor. We got a lot of work to do and to hear! 
Now, it is time to open our present!


Introducing our reprensentative from INFORS HR.. His name is Mr Sharman.. This handsome guy will give us detail explanation about the baby bioreactor in our lab...




Now... It's time to open our present... Tadaaaa~
 

If you haven’t seen Mr. Fermenter before, well.. This is how he looks like. It is already pack like this. Quite handsome right??

Now, let's have a look what Mr Fermenter have...

Part of the fermenter...



Let's get some knowledge about the function of the mr fermenter....

Peristaltic pump >> these pump are removable and autoclavable. silicone tubing connects the inlet side of the pump head to the reagent bottle and the outlet to the 2-way inlet fitting on the vessel top plate.

Drive system >> the motor located within the base unit. the connection to the vessel is simply lifted for removal and replaced for operation.

Base unit >> main housing with a rotameter for controlling gal flow rate, a heater block for temperature control and space for the vessel support frame.

Vessel and support frame >> fitting may vary depending if a bacterial or cell culture system has been specified. stainless steel top plate can be taken away by removing 3 screws.

Control panel >> the 2 LED displays shows the actual values and set point for the various parameters which are measured (C, rpm, pH, PO2, AF and pump)

Reagent bottle and holder >> these are 250 ml capacity and have a steel top plate with two pipes.

Baffle >>To prevent liquid swirl (vortex) and thereby enable the impeller to impose power on the liquid in the form of turbulence and flow and to improve aeration efficiency

Harvest line >> harvesting the desired product.

These are the vessel fittings...



The fitting should include:
An exit gas cooler
A sample device and pipe
A 3-way inlet
An inoculation port
A pH electrode
(pO2, electrode and antifoam and option.)

these cute girl are holding the vessel..this is non jacketed vessel for the fermenter...

Picture below shows the impeller which is Rushton turbine type.. The function of impeller is for the proper mixing and agitating.. There is also the air sparger for air supply..


Picture below shows the condenser.. The function is to cool the liquid in the air outlet resulting in the separation of the foam and other liquid particles..

The rotameter function is to controlling the amount of air entering the vessel... Can be used from
0 l/min to 5 l/min...





THEN, MEDIA PREPARATION....

After knowing all parts of the Mr Fermenter... We need to prepare the media..




Medium is one of the nutrient source for yeast growth. Here we prepare YPG medium which contain of peptone, yeast extract and glucose that is optimum portion for the growing of Saccharomyces cerevisiae.


Here the recipe....
Yeast extract 1 %, Peptone 2%, Glucose 2 %, Distilled water





Mix all the media with distilled water by using the magnetic stirrer.
Then, autoclave it... Picture below shows the media after autoclaving...




What we are going to do actually is.......


huh~ At last.... About 5.00 pm, we have finished the theory class... Little bit tired, but there are such an interesting day knowing the mr fermenter.... Tonight, sleep well... Tomorrow we have another date with Mr Fermenter...~~ (^_^)

assalamualaikum.. hye2 cute gurls, ^^.. I'm intrested with ur post, but I'm lack of information and confidence in differentiate between fermenter and bioreactor.. ok2, my questions now, are your post above related with bioreactor? becoz, I got assignment on 5Litre Bioreactor.. Is it related?

I'm a biotech student from UMP. hope u can share some information about your handsome+awesyem equipment with me ^^..
